Temperature

The ambient temperature is directly related to the quality of the reads. At the time we performed a calibration experiment we had the good fortune that it was a hot week during which the sun would heat up the lab daily. The two graphs below illustrate the temperature as recorded by the genome analyzer and the average intensity of the different fluorophores for each cycle.

There are two conclusions to draw from these two strongly correlating graphs:

  • A higher ambient temperature leads to lower intensities of our measurement and thus to a lower quality of the total run.
  • Temperature variations will affect the experiments.
